Integrated sample prep and qPCR
Combined Sample Prep and RT-qPCR in 12 minutes
The combined Sample Prep and qPCR machine is hosted in the same format as the Minicube Prep
The qPCR is facilitated in another section of the capillary allowing very rapid PCR – we have achieved 40 cycles in less than 7 minutes and we have demonstrated that the RT step can be completed in 2 minutes. Hereto, the sample prep of 1.5 min – gives you 7+2+1.5 = 10.5 min and then additional time for the melting curve to complete a full diagnostics in less than 12 minutes.
All the above steps can be adjusted in the app for full flexibility
A typical protocol looks as follows:
1. Insert 200 ul pipette with sample in the sample orifice
2. Press the “Play button” and the sample is sucked into the device
3. Watch the progress on the device – wait until all 16 LEDs blinks rapidly
4. Examine the results on the App
5. The sample will be internally destroyed so that no amplicons leaks out from the device.
It is possible to make a protocol and retrieve the amplicons, and one should use assays with Uracil-DNA glycosylase (UNG) protection against carry-over.
The device comes with 3 pumps a multi-valve pump with 8 ports with interface to the capillary system and 7 solutions and two peristaltic pumps – one for the sample and one for the waste solution.
The 8 port configuration of the multi-valve pump allows for 1 mastermix and 1 primer mix solution, 2 wash buffers and 1 eluation buffer, the 3 other ports are reserved – 1 for the interface to the capillary which is the pump output and the other two for the cleaning solutions for removing all molecules from the inside of the capillary.
The multivalve pump can be delivered with different configurations e.g.:
A. an extra 6,8 or 12 port valve allowing for up to 11 different primer solutions to be used.
B. an extra 6 port and additionally 5 extra pumps with 6, 8 or 12 port configuration giving maximal 5 x 11 = 55 different primer mixes
C. an extra 12 port and additionally 11 extra pumps with 6, 8 or 12 port configuration giving maximal 11 x 11 = 121 different primer mixes
The two peristaltic pumps comes with standard tubing that needs replacement every 3 months, a tubing is available that only needs replacement every 2nd year.
We provide qPCR kits, and wash solutions and nanoparticles at very low cost, and spare fluid connector parts which also can be purchased directly from the third party suppliers.
STEP 1 : INSERT SAMPLE - USING A 10-200 ul pipette
Insert a pipette with sample into the orifice which always is clean due to the fountain overflow of buffers.
Press the “play icon” on the device and the sample is sucked in. There is no limit in sample size but there is a limit in how big pipette tip the orifice can accept – you can ask for a customized version to take 1-10 ml pipette tips.
STEP 2 : WAIT UNTIL ALL 16 LEDS FLASHES FAST
The machine is now purifying your sample and the progresss can be followed on the 16 LEDs – and when completed they will all start to flash very rapidly.
STEP 3 : READ RESULT ON THE APP
The app reports the qPCR results. Logarithmic curves, qT estimate for internal positive sample control and test result for the given number of targets
STEP 4 : Device is cleaning - LEDs flash randomly
The device is now cleaning wait until the LEDs stop flashing. The device is now clean and you can run another sample. The cleaning takes 1 min and 40 sec.

The temperature control matches that of the Minicube PCR with 20 milli-kelvin accuracy and 5 milli-kelvin precision using NIST calibrated PT1000 sensors for temperature monitoring

The machine is calibrated and deliver a high accuracy and precision in the protocols giving you the same results over and over
speed
With heating rate of 40°C/sec and cooling rate of 20°C/sec you get the fastest speed in the market. 40 cycles can be completed in less than 7 minutes
Higher Speed
We have have shown we can press the system to deliver 40 cycles in 5 minutes, but this is a customized version since it is less mechanical robust. Please inquire if you need the speed
Customized versions available
Number of SOLUTIONS
The standard version comes with 6 ports. Three of these are used for an output port to the capillary and two for the cleaning that removes all molecular traces. This leaves 3 ports for a typical RNA/DNA extraction with “Wash 1”, “Wash 2” and “Eluation buffer”

The multi-valve syringe pump can be expanded to 8 or 12 ports giving access to make much more complicated wash protocols
